Исследование корреляции между экспрессией CD133 на клетках человеческой линии колоректальной аденокарциномы НТ-29 и их устойчивостью к химиотерапевтическим препаратам
В сублиниях HT-29CD133+ и HT-29CD133, полученных после клеточного сортинга, выявлена корреляция между химиорезистентностью и высокой экспрессией СD133. При этом нокаут гена PROM1 (и, следовательно, отсутствие экспрессии CD133) не повышал чувствительность опухолевых клеток к химиопрепаратам, что свидетельствует об отсутствии непосредственного влияния CD133 на формирование химиорезистентности в клетках колоректального рака. Клеточные линии НТ-29 с полным или частичным нокаутом гена PROM1 одинаково чувствительны к ингибиторам протеинкиназ сорафенибу и сунитинибу. Примечательно, что наибольшую устойчивость к ингибиторам mTOR — темсиролимусу и эверолимусу — проявили клетки с полным нокаутом гена PROM1 (KO-HT-29, клон Р1). Полученные данные свидетельствуют о том, что CD133 ассоциирован с химиорезистентностью клеток колоректального рака, но не участвует в её формировании.

Correlational study between expression of CD133 on human colorectal adenocarcinoma cells HT-29 and their resistance to chemotherapeutic drugs
A number of studies have shown the association of CD133 expression with cancer stem cells and with the formation of their chemoresistance. In the present study, in subline HT-29CD133+ and HT-29CD133 obtained after cell sorting, a correlation between chemoresistance and high expression of CD133 was found. On the other hand, the knockout of the PROM1 gene and, as a consequence, the absence of CD133 expression did not increase the sensitivity of tumor cells to chemotherapy, which indicates the absence of a direct effect of CD133 on the formation of chemoresistance in colorectal cancer cells. Different variants of the HT-29 line with complete or partial knockout of the PROM1 gene are equally sensitive to the protein kinase inhibitors sorafenib and sunitinib. Notably, that the highest resistance to mTOR inhibitors, temsirolimus and everolimus, was shown by cells with a complete knockout of the PROM1 gene (KO-HT-29 (P1)). The data obtained indicate that CD133 is associated with the chemoresistance of colorectal cancer cells, but is not involved in its formation.
