A protective role of okadaic acid in liver injury induced by amoxicillin
This study is performed to investigate whether there is a protective effect of okadaic acid (OA) on amoxicillin-induced liver injury of rats. Fifty SD rats (male:female=1:1) were randomly divided into five groups according to body weight and sex. Three groups were treated with 1000, 2000, and 4000 mg/kg amoxicillin daily by intragastric administration. The control group was given carboxymethylcellulose sodium (CMC-Na) solution, and the OA group was given 200 ng/kg of OA by intraperitoneal injection and 2000 mg/kg of amoxicillin by intragastric administration. All rats were fed continuously for 28 days. The relative organ weight index, serum ALT activity and liver MDA content were measured for assessing liver injury. Liver cell swelling, congestion, apoptosis and necrosis were found in amoxicillin-treated groups by liver histopathological examination. The relative organ index of rat liver increased after amoxicillin exposure. Moreover, both serum ALT activity and liver MDA content of amoxicillin-treated groups were higher than that of the control group. However, the degree of these hepatocyte damages was decreased after OA treatment. OA can alleviate the liver injury of rats caused by amoxicillin.
