Perivascular pulmonary artery innervation in human and swine: a comparative study for the development of an experimental model for denervation
Pulmonary artery denervation (PAD) has been proposed as an alternative treatment for pulmonary arterial hypertension (PAH). As a novel method, transcatheter PAD requires extensive investigation in experimental studies, and large animals, such as swine, seem to be suitable regarding vessels diameter and the feasibility of manipulation inside the pulmonary artery (PA). However, data regarding the difference in periarterial innervation between human and swine PA is lacking. The potential reproducibility of experimental findings on PA denervation in clinical settings should be proven. We sought to compare the distribution density and the depth of nerves in perivascular adipose tissue of the trunk and bifurcation of the PA in humans and pigs. An immunohistochemical study was also performed in order to evaluate the differences in neural receptor profiles. There were differences in distribution density and diameters of nerve fibers. However, the depth of nerves in the adventitia and the perivascular tissue did not differ. Immunohistochemical study showed a similar receptor profile in human and swine. The expression of all studied markers in pigs was less pronounced in comparison with humans. Based on the data obtained, it is possible to state that the innervation of the pulmonary artery of a human and a pig share some similarity, and the swine model can be suitable for PA denervation experiments.
