Восстановление метформином β-адренергической сигнализации, активности Аkt-киназы и АМФ-активируемой протеинкиназы в миокарде диабетических крыс
Исследовали влияние метформина (100 и 200 мг/кг в сутки, 4 нед) на стимулирующие аденилатциклазу эффекты β-агонистов и релаксина в миокардиальных мембранах, на активность Akt-киназы, эффекторного звена инсулинового сигналинга, и АМФ-активируемой протеинкиназы (AMPK), энергетического сенсора клеток, в миокарде крыс с диабетом, вызванным высокожировой диетой и стрептозотоцином. Метформин нормализовал соотношение аденилатциклазных эффектов β1/2- и β3-агонистов в миокардиальных мембранах, сниженное при диабете, а также восстанавливал фосфорилирование Akt-киназы по Ser473 и AMPK по Thr172 в миокарде диабетических крыс. Действие метформина было выражено в большей степени в дозе 200 мг/кг. В основе кардиопротективного эффекта метформина лежит его способность восстанавливать адренергическую и инсулиновую регуляцию в кардиомиоцитах и их энергетический статус.

The restoration of the β-adrenergic signaling and the activity of AKT-kinase and AMP-activated protein kinase by metformin in the myocardium of diabetic rats
The metformin (MF) treatment is used to correct diabetic cardiomyopathy in the type 2 diabetes mellitus (DM2). At the same time, the MF effect on the myocardial hormonal systems has been little studied. The aim of the work was to study the effect of 4-week treatment of rats with DM2 induced by high-fat diet and streptozotocin with MF in the doses of 100 and 200 mg/kg/day on the adenylyl cyclase (AC) stimulating effects of β-agonists and relaxin in the myocardial membranes and on the activity of Akt-kinase, an effector component of the insulin signaling, and AMP-activated protein kinase (AMPK), an energy cell sensor, in the myocardium of diabetic rats. The MF normalized the ratio of AC effects of β1/2- and β3-agonists in the myocardial membranes, decreased in DM2, and also restored the phosphorylation of Akt-kinase by Ser473 and AMPK by Thr172 in the myocardium of diabetic rats. The effect of MF in a dose of 200 mg/kg/day was more pronounced. Thus, the cardioprotection action of MF is due to its ability to restore the adrenergic and insulin regulation in cardiomyocytes and their energy status.
