Морфологическая характеристика прогрессии меланомы B16 у мышей линии C57BL/6 с высокой и низкой устойчивостью к гипоксии
Исследовали особенности прогрессии опухолевого роста меланомы B16 у самцов мышей линии C57BL/6 с исходно высокой и низкой устойчивостью к гипоксии. Для оценки чувствительности к гипоксии животных помещали в барокамеру на “высоту” 10 000 м. Через 1 мес после тестирования в барокамере высокоустойчивым и низкоустойчивым к гипоксии животным перевивали меланому B16. Через 19 сут после пассажа опухолевых клеток морфологическими и иммунофлюоресцентными методами оценивали выраженность прогрессии меланомы. Методом ПЦР в реальном времени определяли уровень экспрессии vegf-a и hif-1a в печени интактных мышей и мышей с меланомой. У низкоустойчивых к гипоксии мышей прогрессия опухолевого роста была более выражена, о чём свидетельствуют высокие показатели массы первичного узла опухоли, относительной площади некрозов, показатели пролиферации (митотический индекс и количество Ki-67+-клеток), экспрессия в печени гена vegf-a. У высокоустойчивых к гипоксии животных было выше количество каспаза-3+ клеток, гибнущих по механизму апоптоза. Полученные данные о более выраженных темпах прогрессии меланомы у низкоустойчивых к гипоксии животных следует учитывать при поиске новых прогностических маркеров и методов лечения злокачественных новообразований.

Morphological characteristic of the melanoma B16 progression in C57BL/6 mice with high and low resistance to hypoxia
The features of the B16 melanoma growth progression in male C57Bl/6 mice with initially high and low resistance to hypoxia were studied. To assess the resistance to hypoxia, mice were placed in the hypobaric chamber at a “height” of 10,000 m. One month after testing in the hypobaric chamber, animals with high and low resistance to hypoxia were inoculated with B16 melanoma. Morphological and immunofluorescent methods were used to evaluate the severity of the progression of melanoma in the 19 days after the melanoma passage. The expression level of vegf-a and hif-1a in the liver both of the control group and the mice with melanoma was determined by PCR-RT. Tumor growth progression was more pronounced in hypoxia-resistant mice, as evidence by the high indexes of the primary tumor site’s mass, relative necrosis area, proliferation rates (mitotic index and number of Ki-67+ cells) and expression of vegf-a gene in the liver. Highly resistant to hypoxia animals had a high number of caspase-3-positive cells dying by apoptosis. The obtained results about more pronounced rates of melanoma progression in mice with low resistance to hypoxia ought to be considered when new prognostic markers and methods for treating malignant neoplasms are searching.
