Transcriptomic analysis of skeletal muscle tissue in patients with PICS: Novel mechanisms of pathomorphogenesis and promising therapeutic targets
Skeletal muscle atrophy is one of the hallmark manifestations of persistent inflammation, immunosuppression, and catabolism syndrome (PICS) that hinders patient recovery. Defining the morphological and molecular-genetic changes underlying this process is critical for identifying novel mechanisms of PICS pathomorphogenesis and potential therapeutic targets. In this study, pathomorphological and transcriptomic analyses were performed on muscle biopsies obtained from six patients with high-energy trauma. In patients with PICS, a decrease in muscle fiber cross-sectional area and an increase in the proportion of necrotic fibers were found. A total of 6,471 differentially expressed genes were identified, including genes involved in extracellular matrix remodeling, which represent potential targets for directed therapy. This study presents the first description of the transcriptomic profile of skeletal muscle tissue in patients with PICS. Novel pathogenetic mechanisms and potential therapeutic targets were identified.
