Characteristics of neuropeptide Y-ergic regulation of spontaneous rhythm and contractility in the rat myocardium
We investigated the impact of neuropeptide Y (NPY), both independently and in combination with the selective Y2R antagonist BIIE0246 and Y5R antagonist CGP71683, on the spontaneous rhythm and the amplitude–time parameters of right ventricular myocardial contractility. Intrinsic effects of the selective Y2R and Y5R antagonists on the spontaneous contraction rate of the isolated right atrial preparation with a preserved sinoatrial node were identified. The combined application of NPY and these receptor antagonists reduced the spontaneous contraction of the atrial myocardium. NPY application during Y5 receptor blockade induced a positive inotropic effect, increasing the amplitude of right ventricle myocardial contraction via Y1R activation.
